Size exclusion chromatography (SEC) is one of the key methods used in the characterization of biopharmaceuticals, such as monoclonal antibodies (mAbs). Typical SEC analysis is performed utilizing an LC-UV method that provides the necessary information on aggregates and fragments of mAbs. However, additional detection techniques, such as light scattering and mass spectrometry (MS), can provide more sensitivity and detailed information on the aggregates of intact mAbs, as well as increased detail on the reduced heavy chain/light chain and Fab/Fc fragments of mAbs.

This webinar will provide examples of standard SEC data and compare that to the results achieved with light-scattering detection as part of a multidetector solution. On-line MS analyses with detailed molecular weight results will also be shown for intact, reduced, and digested mAbs—with the addition of data from some smaller polypeptide chains. Optimum results have been achieved by combining these powerful detection techniques with higher resolution, easy to use, 3 μm SEC columns.
